You use KY or Vaseline to lube up the probe so that it slides in easy (Hey, you'd want the same courtesy, right?). Then you very gently nudge it into the anal opening, the key word here being "gently". If it's a female, you'll get at most two or three scales in and won't be able to probe any further. If it's a male, it'll slide in quite a bit further, up to 8 or so scales.
What I did to learn how was I brought my big gal down to my local reptile store, and had them show me how to probe on her, and then I gave it a shot.
The important thing is just to be gentle, and you shouldn't have any problems.
