The process is basically the same as "popping" a male in order to sex it. Only instead of just rolling out the hemipenes you also get the hemipenal casts (which are not actually sperm plugs) and you may get some pre-sperm which cleans the tubes before sperm is released.

Pythons do not create sperm plugs in the traditional sense. And even if they did, you would find them in a female that has been bred, not on the male, which would defeat the purpose of "plugging" anything.
