Scientists and volunteers will search for pygmy blue tongue lizards at Kulpara, north of Port Wakefield this weekend.
The elusive reptile had been thought extinct until 14 years ago.
The team will look for holes and other signs of a lizard population.
Department of Environment wildlife officer Julie Schofield says the number of pygmy blue tongues remains critically low, and it is hoped the survey will help protect them.
"We are finding more populations and getting to know more about it which is fantastic, and to have something that we thought we had lost and to get a second chance to look after it, and keep it around for future generations, is just really great," she said.
