Should I have sphagnum moss as the substrate in it, or nothing, or cypress mulch? And which substrates would need misting, and how much? Thanks.

it definetely needs *something* in it. most of what ive read suggests sphagnum moss, and it works well for me. dont worry so much about how often youre supposed to mist it...it just depends on how dry it is, and youll be able to tell. mine generalyl stays pretty good for a few days without needing another misting

Here's what I do, and I can almost guarantee you'll get a full one-piece shed. I cut an entrance hole in the side of a rubbermaid container, and I place a clump of moist sphagnum in there. This has worked great for me.
