There is no difference. It's how she acts around males that define the difference.
Some girls will even "get into position" if you touch their backs just right.
There is a HUGE difference in how the vulva looks when a rat is in estrus. I have pics but not on my computer and I have no scanner. The vulva will look very swollen and kind of stick out. I will try and get some pics of my rats to post.

I never look at their genitalia when they are breeding LOL... I usually just go off of how they dance around, wiggle their ears, arch their back etc, and take a male 
I keep a cage of females with no males. Be fore I take a female out of that age to breed I check to see who is in estrus. It's better for the ladies if they are in estrus when you put them with a "ready to go" male.

Yup, I've noticed it too. They get all puffy back there... It's more predominate with younger females.
