Is there anything wrong with this?

Soon, I'm going to have to switch my ball python over to rats, since the mice he's eating is too small for him. I feed him one, and after a couple of hours he is looking around for more.

But I don't want to waste these mice! Would it be detrimental at all to feed him two mice per feeding until my supply is gone?

There is absolutely no problem with giving him two, even three mice, depending on his size. If he's big enough to eat a rat, he can definitely handle two mice. Just watch that he doesn't get constipated.

There is no problem with it as long as you think it`s not too much food. Just make sure you feed one at a time if you are feeding live.
