Ok, this may sound really weird, but I have two green Anoles, I have a pretty decent sized one (Jimi)and I check it's under side and it does have those two things next to its anus, then I checked the huge one and it doesn't have them, so I'm really confused, any help at all would be most grateful.

If you are trying to sex your anoles, grab ahold of them gently but firmly and pull the dewlap out from under their chin. The malse dewlaps will extend much farther than the females. The males can come out as far 1/2an inch to 3/4 of an inch. The females will barely pull 1/4 of an inch.

Another couple of less reliable ways to tell are that the males generally have much longer heads. the females heads tend to be much smaller and "pinched" looking.

Yeah, I'm just really unsure, because my really big one doesn't have the things by it's butt, but it looks like a male, and my little smaller one has them but looks like a girl, so I'm really really unsure. And I don't want to hurt them by pulling their dewlaps.

Males will have either a bulge on either side at the base of the tail (if they are mature). Or, enlarged post anal scales that are visible with slight magnification. A mature male will have both.
Some males have relatively small dewlaps and some females have relatively large dewlaps so this method is unreliable.
The animal's size has little to do with sex. Some males are a bit on the small side particularly before they are fully mature and you also have some large females.
The most reliable tests are the two previously mentioned -- the hemipenile bulges and/or the enlarged post anal scales.
