I myself have a royal (ball- as you guys call them) python, he's not aggressive but if i smell of food he will strike. The answer to your first question is no, it doesn't hurt in the slightest. My girlfriend is a little scared because she thinks it will hury so just to prove to her i rubbed the back of my hand with a mouse and he struck, it bled a little but it felt nothing more than a pin prick.
On handling, i would suggest you just go right in there and pick him up, DO NOT hesitate as this shows him you're scared and he'll be able to sense it. If you just go in and gently pick him up, stroke him a little then put him back. This will start to build trust between you and show him your not a threat. Do it a few times and over the course of a week or so you'll notice a big difference. Just put yourself in his 'shoes'. Something comes into your territory and when you threaten it backs off.... this puts him in charge. Also try washing your hands first, if you've touched any other animal that naturally he knows to be afraid of like cats, dogs etc, he'll be able to smell them.
So no it doesn't hurt and to make him stop with the attitude just remember not to hesitate and just go ahead and pick him up. If you show your scared this will encourage him to keep on threatening you. As long as your gentle he'll start to trust you and see you're no threat.
