semen after copulation. The problem you're experiencing is most likely a shedding problem.
When the snake sheds, the entire dermis, ailimentary canal, and hemipenes shed their top layer. We as keepers almost never see the evidence of the internal shed. So either your snake has a slight problem with shedding (I'd boost the humidity slightly) or it has an abnormal discharge that requires a vets attention. Since the animal is otherwise healthy I'd boost the humidity.
If you examine the "plug" under a microscope is there a cell structure or is it an excretion?

I wasn't aware that sperm plugs were only shed after copulation. I've seen MANY cases of sperm plugs shed with the regular shed cycle and also during defecation......not only during a copulation. In ball pythons, you can easily pop out a sperm plug from a sexually mature male that has never been bred.
