Is it safe to assume that a male that is breeding wont have sperm plugs?

Depends what you mean by "Is breeding." I would not expect him to have plugs five minutes after he unhooks with a girl.

But, if he's not hooked up for a day or two, I would expect residual sperm.

I think it's a case-by-case thing. If he's breeding, I would not worry about whether or not he has plugs at any given point in time. Good luck!

I know that sperm plugs can show when a male is sexually mature. Does it have any other significance, though? Like, you're not supposed to clear them out before breeding or anything are you?

I would assume that the plugs fall out when the male everts it's penes, so I would not worry about clearing them out.
