I would start with a Vet check to make sure all is well, then put them both in a small room like a bathroom with some food, toys and the litter box. Would be best if the room doesn't have carpeting. Hopefully this will re-train them and stop the behaviour. You also need to make sure that you clean the area that they have been using and get all the smell out, otherwise the scent triggers them to go there again. Let us know how it goes.
